Need a few extra heads to think about this one for me:

When the bus is cold (i.e. first run of the day) and I use reverse, there is a strange clunk - only happens once and only as it starts rolling.

So, Uni joint, CV, clutch?

Kinda got me stuffed - any and all thoughts appreciated.

Clutch pressure plate is my guess or handbrake shoes releasing from drums...do you park with handbrake on overnight?

Yeah, thought of that. And it could be. But only when I reverse, not drive away, seems sus?
 
Still think it could be your rear shoes, as reverse off one shoe sticking. Which is why reverse only. I,d pull the drums off and give everything a clean, can't hurt, and that way you can inspect your drums and shoes and will know there condition anyway
 
Left it parked with handbrake off - no clunk. Its mostly always the simple things hey!

Now I gotta find time to strip them and see what's wrong!
